Info: Detection of antigen-specific T cells is usually quite difficult due to the little amount of these cells in the peripheral blood (less than 0.1% of lymphocytes). Within this course the multicolour FACS analysis will be used to determine both phenotypic (expression of activation markers) and functional (cytokine secretion) characteristics of such rare cell populations. The primary advantage of this technique is that many distinct measurements can be performed simultaneously on one sample.

Analysis of Proteases by using Biochemical Methods

Info: Cathepsins are proteases of the antigen processing machinery and are crucial in degradation of antigens within the endocytic compartments of antigen presenting cells. These proteases can be regulated by cytokines or viral infection. The specific aim of the practical course is to distinguish levels of cathepsins in primary B cells and dendritic cells after cytokine exposure by biochemical methods, such as western blot and colorimetric substrates.

Functional analysis of the androgen receptor (AR) signalling

Info: The modulation of the AR activity in prostata carcinoma cell lines with AR coactivators shall be studied. Cells are transfected with AR coactivators and receptor activity will be determined in a luciferase reporter gene assay. In addition, intracellular localization of the AR (fusion protein with a fluorescent protein) will be analysed by fluorescence microscopy.

Analysis of androgen receptor protein complexes in prostate carcinoma cell lines
Duration: 2-3 days
Venue: Project Team Spindler, Institute of General Zoologyand Endocrinology
Participants: min. 1, max. 3
Info: The androgen receptor complexes with a variety of proteins. These complexes will be separated by size exclusion chromatography (gel filtration, FPLC). Various fractions will then be analysed by immuno precipitation and immuno blots and additionally also by electrophoretic mobility shift assays.
