Methoden zur Erythrozytenserologie

Method
Antibody elution (chloroform technique)
 
Procedure

1. Wash RBC eight times.
2. Mix 1 vol RBC sediment with 1 vol PBS in glass tubes.
3. Add 2 vol chloroform.
4. Immediately, shake vigorously for 10 sec.
5. Mix gently for another 1 min.
6. Incubate for exactly 5 min at 56°C. 
7. Remove adherant material from the glass tubing with a wooden bar. 
8. Centrifuge the tube 10 min at 2,500 x g.
9. Harvest the aqueous supernatent (eluate) carefully. 

 Verfahren

1. Erythrozyten 8 mal waschen.
2. Im Glasröhrchen 1 Teil Erythrozyten mit 1 Teil PBS mischen.
3. 2 Teile Chloroform zugeben.
4. Sofort kräftig schütteln für 10 Sekunden.
5. Für 1 weitere Minute vorsichtig bewegen.
6. Genau 5 Minuten bei 56°C inkubieren.
7. Mit Holzstäbchen am Glas klebendes Material entfernen.
8. Glasröhrchen 10 Minuten bei 2.500 x g zentrifugieren.
9. Den wässrigen Überstand (Eluat) vorsichtig abnehmen.
